GH pulse simulator
Place each dose, your sleep and your workout on one 24-hour chart and see where the modelled curves and the body's own GH pulses fall.

Modelled from published half-lives; GH response varies by person; not a dosing recommendation. No study has compared bedtime with pre-workout dosing. A clinician decides timing.

- Tesamorelin at 22:00: GHRH-class estimate (borrowed from sermorelin, not measured for tesamorelin) 22:15 to 00:30, overlaps the natural sleep-onset pulse.
- Ipamorelin at 22:00: modelled GH window 22:30 to 22:50, clear of both reference bands.
When does tesamorelin raise growth hormone?
No study has reported when the GH response to a tesamorelin dose peaks or ends. Its labels give blood levels only: peak about 9 minutes after injection, half-life 8 to 38 minutes. For sermorelin, the same GHRH class, GH peaks about 30 minutes after a dose and lasts 2 to 3 hours; the simulator borrows that window as an estimate. (Egrifta SV label; Egrifta WR label; Geref 50 SPC (HPRA))

Is bedtime or pre-workout dosing better?
No human study has compared them on any outcome. The largest GH pulse of the day comes with the first deep sleep, and in men about 70% of daily GH is released in early sleep. Exercise raises GH as well: after a 30-minute bout it peaked 37 to 54 minutes from the start. A clinician decides timing. Why pair tesamorelin with ipamorelin but not sermorelin?
Tesamorelin and sermorelin both act on the GHRH receptor, whose response tops out near 1 µg/kg, so a second one is expected to add little. Ipamorelin acts on the ghrelin receptor instead. In healthy adults a GHRH plus a ghrelin-receptor agonist released more GH than either alone, often more than the sum. Tesamorelin with ipamorelin is untested in people. (Gelato 1984, JCEM; Vance 1984, JCEM; Popovic 1995, JCEM; Tiulpakov 1995, Clin Endocrinol)

How long does CJC-1295 with DAC last?
Its half-life is 5.8 to 8.1 days. In healthy adults one injection raised GH 2- to 10-fold for 6 days or more and IGF-1 1.5- to 3-fold for 9 to 11 days; after several doses IGF-1 stayed above baseline for up to 28 days. GH still came in pulses, with higher troughs between them. (Teichman 2006, JCEM; Ionescu 2006, JCEM)

Where every number comes from
Compounds
- TesamorelinHalf-life 8 min (Egrifta SV) and 11 min (WR) after one dose, 26 to 38 min after 14 days of the original product; time to peak 0.15 h (labels). No study reports when the GH response peaks or ends. (Egrifta SV label; Egrifta WR label; Egrifta label, 2010)
- IpamorelinHalf-life 2 h and GH peak 0.67 h after the start of a 15-min IV infusion in healthy men; no human data under the skin. The curve is a pure decline because the only study was IV. (Gobburu 1999, Pharm Res)
- SermorelinHalf-life 6 to 7 min IV (EU product information) to 11 to 12 min (US prescribing information); time to peak 5 to 20 min under the skin; GH peaks about 30 min (15 to 60) after a dose and lasts 2 to 3 h. (Geref 50 SPC (HPRA))
- CJC-1295 with DACHalf-life 5.8 to 8.1 days; a single dose raised GH 2- to 10-fold for 6 days or more, and pulses kept their timing. Drawn as a weekly dose at steady state, so it is nearly flat across one day. (Teichman 2006, JCEM; Ionescu 2006, JCEM)
- CJC-1295 without DACNo human pharmacokinetic study has been published, so no curve can be drawn from data.
- Sleep-onset pulseThe largest GH pulse of the day comes with the first deep sleep; in men about 70% of daily GH is released in early sleep. Its length varies (1.5 to 3.5 h in the first study). (Takahashi 1968, J Clin Invest; Van Cauter 1998, Sleep)
- ExerciseAfter a 30-min bout, GH peaked 37 to 54 min after the start, higher with harder exercise. (Weltman 2000, J Appl Physiol; Wideman 2002, Sports Med)
