Whey vs Casein: Fast and Slow Protein Differ in the Lab, Not in the Training Trials
Whey spikes muscle protein synthesis faster and higher after a shake. Casein trickles in for hours. Yet in the training trials that compared them over weeks, muscle and strength gains did not differ significantly.

Key takeaways
From 9 sources- After a single drink, whey hydrolysate raised muscle protein synthesis about 93% more than casein at rest and 122% more after exercise in young men1.
- In 48 older men, a 20 g whey serving stimulated muscle protein synthesis more than the same amount of casein2.
- Over 8 to 9 weeks of training, whey versus casein, or fast versus slow blends, showed no significant difference in lean mass or strength gains34.
- In a small 10-week trial in 13 men, 35 g of casein gave similar muscle and strength gains whether taken earlier in the day or before bed5.
- Total daily protein mattered more than type: extra gains from protein supplements stopped near 1.6 g per kg of body weight a day6.

Interactions
- Levodopa
- In people with Parkinson's, high-protein meals and the amino acids leucine, isoleucine or phenylalanine cancelled out levodopa's benefit by competing for transport into the brain. Ask the prescriber how to time protein shakes.
Who should be careful
- High intakes
- No UL exists for protein, but the Food and Nutrition Board advises caution with high intakes from foods and supplements combined. In healthy adults, diets of two to three times the RDA did not appear to harm kidney function or bone health (ODS). A systematic review linked chronic, excessive whey use to kidney and liver effects.
- Milk-derived products
- Whey and casein are proteins isolated from milk (ODS); check the label if you avoid milk products.
- Pregnancy and breastfeeding
- No pregnancy or breastfeeding safety data specific to whey supplements appear in the ODS fact sheet, and LactMed has no whey record. NCCIH notes that many supplements have not been tested in pregnant or nursing women.
- Before you start
- Talk to your pharmacist or clinician before adding this supplement, especially if you take prescription medicines, are pregnant or breastfeeding, or have a chronic health condition.
Fast and slow: what the tracer studies show
Whey and casein are both milk proteins3. In a classic tracer study, whey caused a sharp but short rise in blood amino acids, while casein produced a lower plateau that lasted for hours, probably because it empties from the stomach more slowly7. Whole-body protein synthesis rose 68% after whey and 31% after casein, but casein cut protein breakdown by 34% and gave a more positive net leucine balance over 7 hours7.
Muscle itself responded more to whey in short lab studies. In young men given protein with 10 g of essential amino acids, muscle protein synthesis after whey hydrolysate was about 93% higher than after casein at rest and about 122% higher after resistance exercise; each group had 6 men1. In 48 men around age 74, 20 g of whey raised muscle protein synthesis more than 20 g of casein, which the authors linked to faster digestion and higher leucine2.
Over weeks of training, the gap disappears
Lab spikes measure a few hours. In 16 female college basketball players taking 24 g of whey or casein before and after workouts for 8 weeks, lean mass rose 1.5 kg with whey and 1.4 kg with casein, and no measure differed significantly between groups3.
In 31 resistance-trained men, 20 g of protein after each session for 9 weeks was given as all fast-digesting milk protein (the fraction that includes whey), a 50:50 blend or 80% casein4. The faster drinks raised blood leucine more, yet lean mass and strength rose similarly in all three groups4. The training trials are small, but none found that whey's bigger spike translated into more muscle34.
Casein before bed
Casein's reputation as a bedtime protein comes from overnight studies. In 16 young men who trained in the evening, 40 g of casein taken 30 minutes before sleep was digested and absorbed overnight, raised whole-body protein synthesis and turned net protein balance positive; the rise in muscle protein synthesis, about 22%, was only borderline significant8.
Over 12 weeks of training, 44 young men who took 27.5 g of protein nightly gained more strength and quadriceps size than men given a calorie-free placebo9. Because the placebo had no protein or calories, that trial cannot separate bedtime timing from simply eating more protein9.
A small 10-week trial addressed timing directly: 13 men on diets matched at 1.8 g of protein per kg took 35 g of casein either earlier in the day or shortly before bed, and gains in lean mass, muscle size and strength did not differ5. A supplement maker and a dairy company helped fund it5.
What matters more than the type
In a meta-analysis of 49 training trials, protein supplements added about 2.5 kg to strength gains and 0.3 kg to fat-free mass gains, and extra benefit stopped once total intake reached about 1.6 g per kg a day6. Hitting that daily total is the bigger lever than choosing between two milk proteins6. See how much protein it takes to build muscle and whether the anabolic window is real.
Which one fits you
For the full evidence table, see the whey protein page, and the pea protein versus whey comparison if you avoid dairy.
Pick on habit and preference rather than muscle-building claims:
- Want a quick post-workout shake: whey gives the larger short-term muscle protein synthesis response12.
- Want a slow-digesting protein before a long gap or bedtime: casein releases amino acids over hours78.
- Care most about long-term gains: the training trials found no significant difference between the two345.
- Have a milk allergy: both come from milk, so ask your clinician before using either3.
Quick answers
Common questions
3 answersIs casein better than whey before bed?
Can I mix whey and casein?
Yes, blends were tested: in 31 trained men, all-fast, 50:50 and mostly casein drinks gave similar lean mass and strength gains over 9 weeks4.
