Magnesium for Sleep: What the Glycinate, Threonate and Older-Adult Trials Actually Found

Three small trials in older adults suggest magnesium shortens sleep onset by about 17 minutes. Newer trials of bisglycinate and L-threonate found small benefits. None is large, and the evidence grade is low.

Key takeaways

  • Pooled data from 3 trials in 151 older adults: sleep onset 17.36 minutes faster than placebo; total sleep time up 16 minutes but not statistically significant; evidence quality low to very low [1].
  • In the largest of those trials, 46 older adults took 500 mg magnesium or placebo for 8 weeks; insomnia scores, sleep efficiency and sleep onset improved [2].
  • Magnesium bisglycinate (250 mg elemental magnesium daily) in 155 adults with poor sleep reduced insomnia scores more than placebo after 4 weeks, with a small effect size (d = 0.2) [3].
  • Magnesium L-threonate (1 g/day for 21 days) in 80 adults aged 35 to 55 improved some Oura ring sleep scores and morning mood measures vs placebo [4].
  • Keep supplemental magnesium at or under 350 mg/day unless a clinician advises otherwise [5].

The older-adult trials

A 2021 systematic review found only 3 randomized trials comparing oral magnesium with placebo for insomnia in older adults, with 151 people in total [1]. Magnesium shortened sleep onset by 17.36 minutes (95% CI 7.44 to 27.27); total sleep time rose by 16 minutes, which was not statistically significant [1].

The reviewers rated every trial at moderate to high risk of bias and said the literature is not good enough for confident recommendations, while noting magnesium is cheap and widely available [1].

Magnesium bisglycinate (2025)

In a placebo-controlled trial of 155 adults aged 18 to 65 with self-reported poor sleep, 250 mg of elemental magnesium as bisglycinate reduced Insomnia Severity Index scores by 3.9 points over 4 weeks vs 2.3 points on placebo (p = 0.049) [3].

The authors called the benefit modest. People with lower dietary magnesium intake seemed to respond more, but that was an exploratory analysis [3].

Magnesium L-threonate (2024)

Eighty adults aged 35 to 55 with self-assessed sleep problems took 1 g/day of magnesium L-threonate or placebo for 21 days [4]. Oura ring deep sleep and REM sleep scores improved vs placebo, as did questionnaire measures of mood, energy and alertness [4].

Consumer sleep trackers estimate sleep stages and are not polysomnography, and the trial was short, so treat this as an early signal.

If you try it

Label amounts refer to elemental magnesium. The adult upper limit for magnesium from supplements is 350 mg/day; higher doses commonly cause diarrhea, and people with kidney disease face a higher risk of toxicity [5].

Magnesium binds tetracycline and quinolone antibiotics (take them at least 2 hours before or 4 to 6 hours after magnesium) and reduces absorption of oral bisphosphonates (keep them 2 hours apart) [5]. Our glycinate vs citrate guide covers forms, and the magnesium glycinate monograph covers dosing.

Quick answers

Which form of magnesium is best for sleep?

No head-to-head trial compares forms for sleep. Bisglycinate [3] and L-threonate [4] each have one recent placebo-controlled trial; the older-adult trials used other forms [1].

How long does magnesium take to work for sleep?

The recent trials measured effects at 3 to 4 weeks [3][4]; the older-adult trial ran 8 weeks [2].
